Introduction

 We perform proteomics analysis using an originally developed 2DICAL system and reverse-phase protein array (RPPA).

Our team and what we do

 We have originally developed a new analyzing system of proteome generated by liquid chromatography and mass spectrometry (LCMS), named 2DICAL (2-Dimensional Image Converted Analysis of LCMS). Using this system, we are conducting a discovery of the protein important for medicine and biology, and developing a new integrated system contributing to clinical diagnosis and therapy based on the network information of proteome data. In addition, we have applied our in-house RPPA platform to the discovery and development of biomarkers for companion diagnostics and precision medicine.

Research activities

 We performed proteomic analysis of 626 samples from within and outside the National Cancer Center Research Institute (NCCRI). Three peer reviewed papers were published which we contributed in proteomic analysis. In the paper of Kagami Y. et al.'s "Plk1 phosphorylation of CAP-H2 triggers chromosome condensation by condensin II at the early phase of mitosis" (Sci Rep 2017, 7:5583), we clarified the importance of CAP-H2 Ser288 phosphorylation for condensin II function on mitosis. Our proteomic analysis contributed the identification of the phosphorylation site. In the paper of Yoshimaru T. et al.'s "A-kinase anchoring protein BIG3 coordinates oestrogen signaling in breast cancer cells" (Nat Commun 2017, 8:15427), we clarified the mechanism of the suppressive activity of PHB2 to ERalpha positive breast cancer via the state of BIG3 and PHB2 phosphorylation. Our proteomic analysis contributed to prove the phosphorylationexpression.

Education

 We educate the proteomics analyzing method for young researchers from within and outside the National Cancer Center (NCC).

Future prospects

 We aim to develop a system providing a personalized therapy optimized for the character of each cancer using the proteomic data.
