Introduction

 Research is being conducted to construct evidence for the development of cancer prevention by clarifying the causes of human cancer using a study base of a large-scale cohort study and other studies of local residents.

Research activities

1) Traditional molecular epidemiologic studies to investigate the causes of cancer

 We have conducted several traditional molecular epidemiologic studies to investigate the causes of cancer. By using genetic information obtained from the Japan Public Health Center (JPHC)-based prospective study, we constructed a risk prediction model for upper aero-digestive tract cancer. The 10-year absolute risk for developing upper aero-digestive tract cancer varied by risk factor profile, from 0.09% for the lowest risk group (<50 years of age, non-smoking, and moderate drinking or less) to 6.05% for the highest risk group (≥60 years of age, current smoking, heavy drinking along with Lys alteration in ALDH2). The c-index for the gene-environment interaction model (0.71) was slightly higher than that for the environmental model (0.67). The addition of a gene-environment interaction to a lifestyle-based model might improve the performance to estimate the probability of developing upper aero-digestive tract cancer for Japanese men. Another example of a genetic study is the genome-wide association meta-analysis of pancreatic cancer, in which we reported a new susceptibility locus in the glycoprotein 2 gene.

2) Consortium of large-scale molecular epidemiologic studies in Japan

 We established the Japanese Consortium of Genetic Epidemiology studies (J-CGE), a consortium of large-scale molecular epidemiologic studies in Japan, and have conducted “genome-wide association studies (GWAS) stratified by single nucleotide polymorphisms of the ALDH2 gene” and “meta-GWAS on chronic kidney disease”. Using the results of GWAS on body mass index (BMI) from this consortium, we have performed Mendelian randomization analyses for BMI and colorectal cancer. We reported that BMI is significantly associated with increased colorectal cancer risk.

3) Innovative molecular epidemiologic studies to investigate the causes of cancer

 We are conducting innovative molecular epidemiologic studies to investigate the causes of cancer. One example is the use of somatic genomic information in a cohort study, which may provide novel evidence according to cancer subtype. Another example is the use of gut microflora genomes to discover previously unknown pathogenic bacteria in colorectal neoplasms. We are also preparing a new study to evaluate the emerging risk factor of heated tobacco products.

Education

 We have supervised the research of four graduate students.

Future Prospects

 By analyzing data from the epidemiological studies both within and outside Japan, we hope to contribute to developing evidence for personalized cancer prevention and promoting life-long health.
